On December 13, 1841, a group of congregants, led by Reverend George Higgins, split off from the a Third Baptist Church to form the Second Baptist Church of Southwark. They purchased a lot of land on Fifth Street, below Carpenter, for the erection of a meeting house and burial ground. Following the 1854 Act of Consolidation, the church was renamed to "Calvary Baptist Church" to better reflect its place in the newly consolidated City and County of Philadelphia.

 

By 1875, a Sunday school building had been built on the graveyard located behind the church, and in 1894, the church building was sold and occupied by the Lithuanian Catholic congregation of Saint Anthony's Chapel. Later, Congregation Poale Zedek used the building as their Synagogue. Homes now occupy the site.
